يتقدم عقار جديد طورته شركة دنماركية لينافس أشهر أدوية السمنة، بعدما أظهرت تجارب سريرية حديثة قدرته على تحقيق فقدان وزن أكبر مقارنة بعقار تيرزيباتيد واسع الاستخدام.
وأظهرت نتائج تجربة REIMAGINE 5 أن مرضى السكري من النوع الثاني الذين تلقوا جرعة أسبوعية مقدارها 1 ملغ من عقار كاغريسيما فقدوا في المتوسط 12.4% من أوزانهم خلال 60 أسبوعًا، مقابل 9% لدى من تلقوا جرعة 5 ملغ من تيرزيباتيد، المادة الفعالة في مونجارو وزيبباوند. كما سجل العقار الجديد تحسنًا ملحوظًا في السيطرة على مستويات السكر في الدم.
وتشير شركة نوفو نورديسك إلى أن المقارنة لم تشمل الجرعات الأعلى من تيرزيباتيد، التي قد تصل إلى 10 أو 15 ملغ وتحقق فقدانًا أكبر للوزن. وفي تجربة منفصلة من المرحلة الثالثة، فقد بالغون يعانون السمنة نحو 21% من أوزانهم خلال 68 أسبوعًا عند استخدام كاغريسيما مقارنة بدواء وهمي.
ويجمع العقار بين مادتي سيماغلوتايد وكاغريلينتايد، حيث يحاكي الأول هرمون GLP‑1 المسؤول عن الشبع وإبطاء إفراغ المعدة، بينما يحاكي الثاني تأثير هرمون الأميلين المرتبط بتنظيم الشهية ومستويات السكر. وتعد سيماغلوتايد المادة الفعالة في أوزمبيك وويغوفي.
وقال الرئيس العلمي للشركة مارتن هولست لانج: إن النتائج مشجعة وتظهر فعالية قوية في علاج السمنة والسكري. ولم يحصل العقار بعد على موافقة إدارة الغذاء والدواء الأمريكية، لكن الشركة تقدمت بطلب رسمي وتتوقع صدور القرار خلال العام الجاري، مع خطط لطرحه في الأسواق مطلع العام المقبل.
وشملت أبرز الآثار الجانبية أعراضًا هضمية مثل الغثيان والتقيؤ والإسهال والإمساك وآلام البطن، وهي شائعة في أدوية فئة GLP‑1. وتتنافس نوفو نورديسك مع شركات أخرى أبرزها إيلي ليلي، وسط ارتفاع عالمي في الطلب على علاجات فقدان الوزن والسيطرة على الشهية.
A new drug developed by a Danish company is set to compete with the most famous obesity medications, after recent clinical trials showed its ability to achieve greater weight loss compared to the widely used drug terzepatide.
The results of the REIMAGINE 5 trial indicated that patients with type 2 diabetes who received a weekly dose of 1 mg of the drug cagrizema lost an average of 12.4% of their weight over 60 weeks, compared to 9% for those who received a 5 mg dose of terzepatide, the active ingredient in Monjaro and Zepbound. The new drug also showed a significant improvement in controlling blood sugar levels.
Novo Nordisk notes that the comparison did not include higher doses of terzepatide, which can reach 10 or 15 mg and achieve greater weight loss. In a separate phase three trial, adults suffering from obesity lost about 21% of their weight over 68 weeks when using cagrizema compared to a placebo.
The drug combines the substances semaglutide and cagrilintide, with the former mimicking the GLP-1 hormone responsible for satiety and slowing gastric emptying, while the latter mimics the effects of the amylin hormone associated with appetite regulation and blood sugar levels. Semaglutide is the active ingredient in Ozempic and Wegovy.
Martin Holst Lange, the company's scientific president, stated that the results are encouraging and demonstrate strong efficacy in treating obesity and diabetes. The drug has not yet received approval from the U.S. Food and Drug Administration, but the company has submitted an official application and expects a decision to be made within this year, with plans to launch it in the markets early next year.
The most notable side effects included gastrointestinal symptoms such as nausea, vomiting, diarrhea, constipation, and abdominal pain, which are common in GLP-1 class medications. Novo Nordisk is competing with other companies, most notably Eli Lilly, amid a global increase in demand for weight loss and appetite control treatments.
